I think it would be better for you to make it up as you go along. Why? Because it would give you an opportunity to find what you enjoy most and are best at. And who knows? Maybe it could still be a successful experiment like Megatokyo.
I think the quarterstone of your style as far as comics have been concerned, have been short little querky strips about events that people can relate to, through frustration or otherwise.
Having a dedicated ongoing plot based comic would be nice, but again, it would tie you down too much, and tie the fans down too much expecting more from you.
The question then becomes: What do you find you enjoy more? Thinking out your comic, or making it as you go along. Or, just making a comic when a random idea pops its head out?
I seriously don't know what I'd like, especially since I read all three types. However it isn't too common you'll find the second type.
In other words, choose what will be more valuable for you time-wise.
